There are a couple of really long driving days on this trip, especially from Denver to Custer and then from Custer to Cody. The trip has been broken up as much as possible, with a couple of two- or three-night stopovers in some locations, so you'll have plenty of time to rest.
You'll be travelling in a van with a group of travellers, so conditions may feel a little cramped. This is all part of the adventure, however, and you might even make some new friends.
As you'll be travelling through cattle ranch country, there will be a lot of meat served in these regions (beef and bison). Vegetarians may find meal selection limited.
A lot of the places you visit in South Dakota are in prairie country, so expect warm days but cool mornings and evenings. It's a good idea to use sun protection and drink lots of water during the days, and bring warm clothing for the nights.
